Seems to be very weird issue. Facing the same issue on my machine as well. Well I would suggest of removing the update panel for now and do a Postback :P

I think the issue is with the master page not able to load. Can you check if your master page is available under _catalogs\masterpage folder through browser or SPD after the deployment?

I think, you might have missed the reference to jQuery script file. Either add the latest jQuery script file in the master page or script tag just above you add $(document).ready(...) stuff with src.

I think the issue is with the web part. Have removed the webpart and recreated(without any code changes) and shockingly it is working fine
