Is it possible to create a webpart for document library that will look like this?

+1. To add: 1) You can use document library view with grouping by the calculated column, without creating a custom webpart. 2) Probably, it would be better to use =UPPER(LEFT([Title],1)) formula, to ensure that all the letters are uppercase. –  Andrey Markeev Nov 11 '11 at 5:39

I used this jquery library recently to create a similar effect. Very easy to use. I simply created a DVWP that output my content in the format I wanted an in an unordered list.

Then using this, it creates the alphapet list automagically:
